Alanine Aminotransferase (ALT) reagent measures ALT concentration in heparinized plasma by a kinetic rate method. The Synchron system automatically proportions the sample and reagent volumes into the cuvette at a ratio 1:11. ALT catalyzes the reversible transamination of L-alanine and alpha-ketoglutarate to pyruvate and L-glutamate. Pyruvate is then reduced to lactate in the presence of lactate dehydrogenase with concurrent oxidation of reduced Nicotinamide adenine dinucleotide (NADH) to NAD. The change in absorbance at 520 nm is directly proportional to ALT in the sample, and is used by the system to calculate and express ALT concentration.
