Used for the determination of alpha glutathione S-Transferase (αGST) concentration in human serum using the Kamiya Biomedical Human Alpha GST ELISA kit (KT-629).

Principle:

This assay employs the solid phase sandwich ELISA technique. A monoclonal antibody specific for αGST is pre-coated onto the microplate. Standards, controls, and diluted samples are pipetted into the wells and any αGST present is bound by the immobilized antibody. After incubation for 60 minutes on a shaker, the plate is washed to remove unbound substances. Conjugate is added to each well, followed by a 30 minute incubation. The plate is washed again, substrate is added to each well, and colour develops in proportion to the amount of αGST in each sample. Following a 15 minute incubation, an acid solution is added to stop colour development. The wells are read on a microplate reader at 450nm with a correction wavelength of 630nm.
